白粉病胁迫下甜瓜叶片蛋白组学分析Proteomics Analysis of Melon Leaves under Powdery Mildew Stress

摘要(Abstract):
为深入研究甜瓜抗白粉病的分子机制,本文在蛋白质水平上探讨甜瓜应对白粉病胁迫的可能机理,寻找特定的白粉病胁迫应激反应蛋白,为分离、鉴定和克隆抗白粉病的基因奠定基础.本研究采用蛋白质双向电泳技术(twodimensional gel electrophoresis,2-DE),对甜瓜白粉病的感病品种"JS"和抗病品种"MR-1"叶片中差异超过1.5倍的36个蛋白质点进行MALDI-TOF-TOF/MS分析和数据库检索.鉴定出31个差异蛋白质,将鉴定出的可能与甜瓜白粉病相关的差异蛋白进行了初步分析.结果表明:白粉病诱导处理对两种甜瓜的叶片蛋白质表达有明显的影响,两种甜瓜的电泳图谱间存在较为明显的差异.相比而言",MR-1"中表达量下调的蛋白质点数要明显少于"JS",而表达量上调和特异性表达的蛋白质点则明显多于"JS",这种表达差异的存在可能是"MR-1"具有较强抗性的一种重要生理响应.
